Diverse populations, like the United States, lack accurate data on frequencies of lactase persistence (LP). This is due, in part, to the multiple SNPs that can indicate lactase persistency in populations with different ancestry. This project proposes a two-part research project that includes a method development and a genotyping phase. Creation of multiple qPCR assays inclusive of diverse SNPs would facilitate the data collection process that is currently dominated by the more labor-intensive and time-consuming PCR-RPLF technique. Analysis of a diverse representative sample population would provide not only a more accurate estimate of lactase persistence frequencies, but also information on which SNPs contribute to LP frequencies in the U.S.. This information could help update U.S. dietary guidelines to reflect that a significant part of its diverse population is lactase nonpersistent and hence experiencing adverse gastrointestinal symptoms after consuming lactose-containing products.
